Accelerating and De‑Risking Inhaled Drug Development with In Silico Tools

Abstract:

This article highlights the growing role of mechanistic in silico modeling in addressing long-standing challenges in inhaled drug development, particularly the disconnect between in vitro measurements and in vivo performance. By linking single particle benchtop aerosol characterization with computational simulations, it becomes possible to track the evolution of an aerosol from emission through to its deposition in the lungs, providing a more physiologically relevant and predictive understanding of particle behavior. This approach generates validated, high-quality input data for simulations such as pulmonary deposition and physiologically based pharmacokinetic (PBPK) models, improving predictive accuracy and helping to reduce risk in the development of inhaled therapies.
