Abstract:
Dry powder intranasal delivery is a rapidly emerging area that is applicable to a diverse range of therapeutic approaches, including mucosal vaccination, rapid systemic delivery, and central nervous system (CNS) treatment via the olfactory nerve-to-brain route. While nasal sprays have been extensively used for decades, dry powder intranasal products can provide many advantages similar to those of pulmonary dry powder inhalers, including shelf stability without refrigeration, an efficient delivery profile, and ease of use for patients. Nasal dry powders can be formulated for a wide range of active pharmaceutical ingredients (APIs), including small molecules, proteins, peptides and nucleic acids, etc. This work focuses on formulation approaches for biologic APIs, with case studies for a protein, a peptide and a nucleic acid in lipid nanoparticles described. Together, the three case studies show that proof-of-concept intranasal dry powder formulations with ambient-temperature shelf stability can be achieved across diverse biologic APIs.
