Cellular and Subcellular Drug Trafficking in Bronchial Epithelial Cells: Mechanisms Controlling Drug Disposition

Abstract:

A series of in vitro studies aimed to gain an understanding of the mechanisms that might promote the retention of inhaled drugs in the lung tissue by investigating the disposition of five marketed bronchodilators and one investigational compound in the human bronchial epithelial cell line, Calu-3. The results suggest that lysosomal sequestration in epithelial cells and potentially, an efflux system present at the air-epithelium interface, limit pulmonary drug absorption. Those cellular and subcellular processes might therefore prolong the residence time of inhaled drugs in the lung by creating a drug depot within the airway epithelium.
