Abstract:
The effect of temperature cycling under controlled and realistic environmental conditions on unprimed and subsequent delivered doses of commercially available metered dose inhalers (MDIs) was investigated. The inclusion of ethanol in a solution-based formulation appeared to inhibit loss of prime. Drainage of propellant from the metering chamber of suspension MDIs appeared to leave API residue, causing a reduced unprimed delivered dose and increase in subsequent doses which may influence product efficacy and safety and creates a requirement for wasteful priming doses.
