Abstract:
A novel process for creating homogeneous ordered mixtures of micronized drug and small porous carrier particles is described. The process involves spray drying a co-suspension of micron-sized particles dispersed in a liquid non-solvent. Strong adhesive forces between the drug particles and carrier particles are anticipated, with the low-density ordered mixtures forming “respirable agglomerates’” with an aerodynamic size less than 5 μm. The engineered particles exhibit significant improvements in lung targeting and dose consistency relative to spheronized particles and conventional blends of micronized drug and coarse lactose.
